Can you clarify me on this 5th (5-methylcytosine)and 6th nucleotide (5-hydroxymethylcytosine)?

Are they strictly considered the 5th and 6th nucleotide? I haven't had time to read the paper, but are these nucleotides considered reversible? Thank you.
